المستخلص: Background: Preptin hormone is composed 34- amino acids (MW 4030 Da) peptide hormone that is secreted with insulin and amylin from the pancreatic β-cells. Oxidative stress is an imbalance in the levels of pro-oxidants and antioxidants in which the levels of pro-oxidants are in excess. The Aim of Study: To evaluate the role of serum level of preptin hormone in children with obesity. Materials and Methods: In this study was carried out in Salah Al-din General Hospital in Tikrit City, in the period from December 2017 to April 2018, during this period a total of 80 children were included in this study. The children were divided into two groups; group one constituted: 43 obese children (23 males, 20 females), while group two was composed of 37 apparently healthy normal weight children (19 males, 18 females). Their age were from 5 to 14 years. A venous blood samples were collected from all the children included in this study and the sera were separated from the blood samples by centrifuge for the estimation of serum levels of preptin hormone were measured via ELISA, blood Glucose, Lipid profile, Glutathione peroxidase, Catalase, Malondialdehyde, Blood urea, Albumin, Total protein and Globulin in were measured via UV-VIS Spectrophotometer. Results: It was found in this study that there was a significant difference (P= 0.0004) in the concentrations of serum level of preptin hormone in obese children (118.7 ± 44.9 pg/ml) compared with the control (27.6 ± 12.9 pg/ml), (P= 0.0003) in the concentrations serum level MDA in obese children (3.5 ± 0.8 μmol/L) compared with the control (1.7 ± 0.5 μmol/L), (P= 0.0005)in the concentrations serum level GPx in obese children (3.22 ± 0.9 U/L) compared with the control (12.53 ± 3.0 U/L), (P= 0.0005) in the concentrations serum level CAT in obese children(19.69 ± 4.11 K/ml) compared with the control(47.00 ± 7.82 K/ml), (P= 0.05) in the concentrations of serum level of TC in obese children(170.7 ± 26.6 mg/dl) compared with the control (159.1 ± 16.3 mg/dl), (P= 0.044) in the concentrations of serum level of HDL in obese children (48.66 ± 5.60 mg/dl) compared with the control (45.26 ± 6.62 mg/dl) and (P= 0.0002)in the concentrations of serum level of Blood urea in obese children (33.90 ± 6.77 mg/dl) compared with the control (28.07 ± 3.8 mg/dl). There was a non-significant difference regarding the serum levels of TG, LDL, VLDL, TP, Alb, Glob and F.B.S between the obese children and the control (P ˃ 0.05). The correlation between serum level of preptin hormone and other biochemical parameters has appeared that serum preptin there was a significant positive correlation with BMI and MDA. While there was a significant negative correlation between the serum level of preptin hormone with HDL, Alb, CAT and GPx. There was no correlation between preptin hormone concentration with concentrations of (total protein, globulin, TC, TG, LDL, VLDL, blood urea, and glucose). Conclusion: The serum level of preptin was found to be a significantly higher in obese children compared with control, with a significant increase with age and a more significant increase in females compared males.
