Water Microbial diversity is huge and everybody deals with it directly every day. The major question is, how can we deal enough with water safety to remove the organic and inorganic contaminates because it is directly related to the health problem. Here the main challenge is to know the biological diversity of water samples with the help of metagenomics techniques. Metagenomics is a powerful method to study the bacterial diversity in environmental samples without using any cultural techniques. It provides a broader knowledge of bacterial diversity and avoid any risks. Till now only 1% bacterial species are cultivated .but with the help of unculturable techniques we can study rest of bacterial species 99%. This study was conducted on 7 water samples collected from Tripoli (Libya) including waste water treatment station. The genoms of microbial content in those samples were extracted by DNA isolation kit and 16S rRNA nucleotide sequencing was performed by Sanger shotgun sequencing and analyze by bioinformatic tools. The results of this study identified 7 new strains that included 05 families: Vibrio, Staphlococcus, Aeromonas, pseudomonas, Bacillus. Mostly identified bacteria were pathogenic to humans. This study will help in reducing the water borne diseases which are to using several emerging bacterial pathogens.
