Influenza A (H3N2) viruses have been slowly changing over the past few years in terms of binding to various types of RBCs as well as growing poorly in MDCK cells and embryonated eggs. This project investigated some of these viruses using a variety of assays such as receptor binding assays, virus copy number, neuraminidase activity and TCID50. In addition, viruses were passaged in MDCK cells and MDCK-SIAT1 cells to determine their growth properties. Recent H3N2 viruses gave variable results in terms of their haemagglutination ability with different RBC types. Receptor destroying enzyme and sodium metaperiodate binding assays also showed some differences in the binding specificity among the viruses tested. MDCK-SIAT1 cells were better than MDCK cells in growing H3N2 viruses and guinea pig RBCs gave the highest haemagglutination titers.
