Effect medium volume and explants density on in vitro shoot formation and growth of pineapple " Ananas comosus L. Mer " cv. Moris

Abstract: The effect of combinations of four explants density (one, two, three and four explants) and ten volumes (3, 4, 5, 6, 7, 8, 9, 10, 11, 12 ml) of full strength liquid MS medium enriched with sucrose at 20 g/l and 6-benzyalaminopurine (BAP) at 2.0 mg/l on the Moris pineapple in vitro shoot formation rate, shoot length and expected total shoots per one liter of medium were investigated. The results showed that a difference of 2 to 5 ml in the medium volume per culture could cause a loss of 50 % of the possibly obtainable rate of shoot formation. At density of one explant per culture, 10 ml of medium resulted in formation of 13 shoots while 4 ml of medium resulted in formation of 6 shoots per explant. At density of two, three and four explants, using of 11, 9 and 10 ml of medium per culture resulted in formation of 10, 11 and 7 shoots per explant respectively while using of 6, 7 and 5 ml of medium per culture resulted in formation of 5, 5 and 3 shoots per explant. Overall, the shoots formation declined from 9 at density of one to 4 shoots per explant at density of four. The medium volume per culture had no independent significant effect on shoot formation per explant but interacted significantly with explants density. Lower explants density (one explants) and larger medium volume (10 ml) was best treatment for shoot formation per explants (13 shoots). However, the highest total shoots (7500 shoots) per liter of medium obtained using three explants and dispensing the medium at smallest volume (4 ml) per culture.
