Phytochemical Analysis and Antimicrobial Activity of Crude Extracts Obtained from Phillyrea latifolia L. Collected from Jordan

المستخلص: Phillyrea latifolia L. is a species of the Oleaceae family. It is a medicinal shrub, native to the Mediterranean forest, landscaping purposes. This study aims to assess the antimicrobial activity, antioxidant activity and phytochemicals analysis of Phillyrea latifolia L. in Jordan. Antimicrobial activity of ethanolic and methanolic extracts of Phillyrea latifolia L. were evaluated against eight Gram negative bacterial species [Pseudomonas aeruginosa (ATCC 27853), Pseudomonas aeruginosa (Clinical isolate), Serratia marcescens (ATCC 27117), Serratia marcescens (Clinical isolate), Escherichia coli (ATTC 25922), Escherichia coli (Clinical isolate), Proteus vulgaris (ATCC 29905) and Klebsiella pneumonia (Clinical isolate)], and five Gram positive bacterial species [Staphylococcus aureus (ATCC 29213) Staphylococcus aureus (Clinical isolate), Coagulase-Negative Staphylococci (Clinical isolate), Bacillus cereus (ATCC 11778) and Micrococcus luteus (ATCC 9341)] and two fungal species (Candida albicans and Aspergillus niger) using agar well diffusion method and broth dilution method. Results showed that ethanolic extract was more effective against most studied bacterial species than methanolic extract. The maximum inhibition zone (25 mm) was obtained by using Phillyrea latifolia L. ethanolic extract against Coagulase-Negative Staphylococci (Clinical isolate), and the minimum value (11 mm) was obtained by using Phillyrea latifolia L. methanolic extract against Staphylococcus aureus (Clinical isolate). No activity for ethanolic and methanolic extracts of Phillyrea latifolia L. against fungal species. The minimum inhibitory concentration (MIC) of Phillyrea latifolia L. were measured for tested microorganisms, and results showed that ethanolic extract of Phillyrea latifolia L. had the lowest MIC value (24 mg/mL) against Coagulase-Negative Staphylococci (Clinical isolate), while the highest MIC value (90.90 mg/mL) for methanolic extract of Phillyrea latifolia L. was found against staphylococcus aureus (Clinical isolate). Furthermore, the stable free radical 2, 2-diphyenyl-picrylhydrazyl (DPPH) method was used to evaluate the antioxidant activity of Phillyrea latifolia L. extracts. Results showed that the highest rate of radical scavenging activity (92%) was obtained from ethanolic extract and so the lowest rate of radical scavenging activity was recorded for ethanolic extract (56%).On the other hand, Phytochemical analysis carried out in Phillyrea latifolia L. revealed the presence of flavonoid, alkaloids, glycosids, tannins, steroids, phenols and saponins in methanolic extract. Ethanolic extract of Phillyrea latifolia L. showed flavonoid, alkaloids, glycosids, tannins, steroids and phenols, but the absence of saponins.
